    摘要: A method for determining the amount of carbohydrate deficient transferrin (CDT) in blood serum involves saturating a sample of the blood serum with iron to provide isoforms of transferrin, separating the isoforms of transferrin from other serum proteins in the sample, and developing a chromatogram which relates the separated isoforms with different pI values. The separation step is preferably carried out in an ion exchange column using a salt gradient and the chromatogram is preferably developed using a high performance liquid chromatograph (HPLC). The method is particularly useful for identifying heavy alcohol consumption in a person, which is related to isoforms of transferrin having pI values of 5.9 and 5.7.

    摘要: Disclosed is a method for detecting and quantitating oligonucleotides with charged internucleotide linkages in biological fluids. In this method, a biological fluid sample is contacted with an anion exchange resin at from 40.degree. C. to 65.degree. C. for a time sufficient to enable oligonucleotides in the sample to adsorb to the resin. The absorbed oligonucleotides are then desorbed with a buffer having a salt concentration of about 1 M to 2.5 M and a pH in the range of about 6.5 to 7.5, the desorption being performed at about 40.degree.-65.degree. C. The oligonucleotides so released are then detected and quantitated.

    摘要: An easily-assembled, heat-resistant connector for releasably joining end portions of two capillary tubes in end-to-end fashion for use in chromatography without interrupting fluid flow or interfering with chromatographic results, comprises a cylindrical sleeve which has a bore which tapers toward a center point to form press-fit seals with the tips of two capillary tubes. A first ferrule is mounted on the end portion of a first capillary tube, and a second ferrule is mounted on the end portion of a second capillary tube. A jacket surrounds and contains the sleeve, and a first adjustment screw mounted on the jacket pushes against the first ferrule to deform the ferrule into sealing contact with the outside surface of the first capillary tube and with the inside surface of the sleeve. A second adjustment screw mounted on the jacket pushes against the second ferrule to deform the ferrule into sealing contact with the outside surface of the second capillary tube and with the inside surface of the sleeve. The pressure on the first ferrule may be adjusted individually and independently of the pressure on the second ferrule and the pressure on the second ferrule, may be adjusted individually and independently of the pressure on the first ferrule. The pressure from the ferrules also hold the press-fit seals in sealing contact.

The valve has a valve seat (12) with an outlet channel opening (18) located in its center and a valve body (13) that cooperates with the valve seat. The valve body (13) can be moved with the aid of a piezo transducer, actively in the opening direction and, while being monitored, actively in the closing direction. The opposed faces of the valve seat (12) and the valve body (13) are so shaped that an annular gap (16) that widens radially towards the outside starting from the opening (18) of the outlet channel is produced. When the valve is in the closed position, this results in the formation of a narrow sealing zone directly at the edge of the opening of the outlet channel.

    摘要: A sample of a hydrocarbon oil containing asphaltenes is chromatographically analyzed by forming a mixture of the oil with a weak solvent. The mixture is passed in contact with a column of a stationary phase of fine solid particles of fully functionalized material, followed by a weak solvent. The solvent, after recovery from the column, is analyzed for aromatics by UV-absorption of UV radiation in the range 200 to 400 nm. The absorbance of the UV light by the irradiated eluents across the UV wavelength range is monitored and the integral of absorbance is derived as a function of photon energy across the wavelength range. The magnitude of the derived integral in at least one time interval corresponding with aromatics in the eluent from the stationary phase is measured as an indication of the level of aromatics in the oil sample. The weak solvent may be followed by a strong solvent which, in turn, may be followed by a strong solvent which is modified by the addition of a hydrogen bonding solvent.

    摘要: A valve arrangement for controlling the flow of a sample of blood gases along a capillaric streamway from a probe towards an analyzing apparatus includes a body 12 having an inlet 36 for control fluid and a reciprocable valve head 20 movable from a first position in which inlet 36 is isolated from a second inlet 30 for carrier gas and blood gases towards a second position in which inlets 36, 30 are in communication. The arrangement is such, that in the first position of the valve head 20 the carrier gas and blood gases can flow from inlet 30 towards an outlet 32. However, in the second position the pressure of the control fluid prevents the flow of the carrier gas and blood gases through the inlet 30.
