The method comprises culturing a yeast host cell comprising: (1) a first DNA molecule comprising a DNA sequence encoding P4H&agr; subunit operably linked to a promoter functional in the yeast host cell, (2) a second DNA molecule comprising a DNA sequence encoding P4H&agr; subunit operably linked to a promoter functional in the yeast host cell, and (3) a third DNA molecule comprising a DNA sequence encoding a polypeptide or peptide operably linked to a promoter functional in the yeast host cell, wherein the polypeptide or peptide is one which, when hydroxylated, forms the hydroxylated triple helical protein; and wherein during culturing, each of the first DNA molecule, the second DNA molecule and the third DNA molecule are replicated, stably retained and segregated.

The invention further provides a biocompatible prepolymer composition comprising hydrophilic and hydrophobic prepolymers, wherein at least one of the hydrophobic prepolymers has at least one ethylenically unsaturated functional group and at least 5% of the prepolymers have two or more ethylenically unsaturated functional groups and water. Where the cells an/or suitable progenitor cells thereof are chondrocytes, embryonic stem cells and/or bone marrow stromal cells, the methods of the invention are suitable for treating, for example, articular cartilage degeneration associated with primary osteoarthritis. Also disclosed is a device having tissue-like characteristics for treating diseased or damaged tissue in a subject, wherein the device comprises cells of a type(s) normally found in healthy tissue corresponding to the diseased or damaged tissue, and/or suitable progenitor cells thereof, in association with bioresorbable beads or particles and optionally a gel and/or gel-forming substance.
