Abstract: A system that incorporates teachings of the subject disclosure may include, for example, a process that includes obtaining a porous medium comprising a porous material having a first shape and an initial porosity profile. The porous medium is engaged with a cavity in a fluidic device, wherein the cavity is in fluid communication with a channel of the fluidic device. The first shape of the porous material can be adjusted to a second shape resulting in the initial porosity profile being adjusted to a target porosity profile. Such adjustment can be accomplished by the engaging of the porous medium with the cavity, by pre-adjusting a shape of the porous media before insertion into the cavity, or by some combination thereof. Other embodiments are disclosed.

Abstract: The present invention provides methods for enhancing chemical reactions of molecules, e.g., biomolecules, with destructible surfactants. The chemical reactions may involve and/or be associate with analysis, e.g., solubilizing, separating, purifying and/or characterizing the molecules. In one aspect, the anionic surfactants of the present invention may be selectively broken up at relatively low pH. The resulting breakdown products of the surfactants may be removed from the molecule/sample with relative ease. The invention has applicability in a variety of analytical techniques.

Abstract: The present technology is directed to methods of preparing affinity chromatographic columns using streptavidin-conjugated particles and biotinylated affinity agents (i.e., biotinylated oligonucleotides, biotinylated antibodies or biotinylated antigen-binding fragments thereof). The methods of the present technology can be utilized to perform affinity capture assays in a high-throughput, efficient manner. Further, the affinity chromatographic columns disclosed herein are customizable due to the ability to utilize any biotinylated affinity agent. The methods herein utilize an on-line loading (i.e., loading the column with biotinylated affinity agent via a liquid chromatography system).

Abstract: Described is a method for determining a dwell volume of a liquid chromatography system and a liquid chromatography system that can determined the system dwell volume. The method includes mixing a flow of a first solvent with a flow of a second solvent to form a solvent mixture. The flows of the first and second solvents are decreased and increased, respectively, to generate a gradient composition. A system pressure of the liquid chromatography system is measured to determine a pressure trace defined as the measured system pressure as a function of time. The dwell volume of the system is determined from a time delay determined between the gradient composition at the mixing location and the pressure trace. The method can be performed with a liquid chromatography system having a chromatographic column or a flow restrictor used in place of the chromatographic column.

Abstract: Exemplary embodiments are directed to methods and systems for controlling fluid parameters within a detector. A first and second fluid pump manager control the flow of a first and second fluid to one or more heating/cooling devices, and a mixer receives and mixes the first and second fluids. An optical detector flow cell receives the fluid mixture from the mixer, and a pressure regulator controls the pressure at the optical detector flow cell. Thus, the composition, temperature, and pressure of a fluid mixture entering an optical detector flow cell can be controlled in real time.
