The invention is directed to a method and device for simultaneously testing a sample for the presence, absence, and/or amounts of one or more of a plurality of selected analytes. The invention includes, in one aspect, a device for detecting or quantitating a plurality of different analytes in a liquid sample. Each chamber may include an analyte-specific reagent effective to react with a selected analyte that may be present in the sample, and detection means for detecting the signal. Also disclosed are methods utilizing the device.
Abstract:
A method for making a test device is provided. The method can comprise providing (a) a substrate comprising a plurality of wells, wherein each well comprises at least one dimension no greater than 1 mm; (ii) a robotic dispensing system comprising at least one piezoelectric dispenser; and (iii) a liquid solution comprising at least one analyte-specific reagent. The method further comprises delivering a volume of the liquid solution into at least some of the wells using the at least one piezoelectric dispenser. The volume can be no greater than about 1 microliter. The method can further comprise placing a cover over the wells.

A multicompartment assay device comprising at least one compartment comprising a viable organism control medium capable of sustaining growth of total microbial organisms; at least one compartment comprising a medium capable of selectively sustaining growth of target microbial organisms; and, at least one compartment comprising an antimicrobial susceptibility interpretation medium. Also, a method of detecting the presence of target microbial microorganisms in a biological sample and of simultaneously determining the susceptibility of such microorganisms to antimicrobial agents by use of such a device.

Methods for detecting and treating polymicrobial infections, wherein a mixed population of microbes (e.g., bacteria) are present in a patient sample and the microbes are not first isolated from the sample. For example, the present invention describes specific polymicrobial infections and methods of treating said infections, wherein a particular antibiotic or a group of antibiotics are selected based on the composition of the polymicrobial infections.
Abstract:
A method for producing tissue constructs comprising two or more distinct regions, each of which comprises a different cell population or lineage is described. The method involves providing a support substrate or substrate assembly comprising two or more physically distinct regions, wherein the two or more physically distinct regions of the support substrate or substrate assembly are different from each other; and depositing/positioning one or more cells on the support substrate or substrate assembly. The cells can form a continuous monolayer with at least two zones, e.g., a proliferative zone and a nonproliferative zone, that can act as in vitro intestinal models. The models are two-dimensional, thus facilitating rapid and facile imaging. Systems comprising the tissue constructs and methods of using the constructs to study the effects of pharmaceuticals, nutraceuticals, and metabolites on intestinal cells are also described.

The present invention provides a device for detecting aerobic bacteria, anaerobic bacteria and fungi in a targeted material which comprises of a plurality sided slide, and a vial, wherein a first side of said plurality sided slide is coated with a first predetermined medium for aerobic bacteria growth and a second side of said plurality sided slide is coated with a second predetermined medium for fungal (mold and yeast) growth, and wherein at the bottom of said vial a third predetermined medium is embedded to determine a level of presence of anaerobic bacteria.

A multi-compartment resealable container for testing for the presence of micro-organisms is provided with or adapted to receive a sample in a first substantially rigid transparent compartment, a growth medium in a second compartment, and a sanitizer in a third compartment, the compartments being separated by foil seals, the second and third compartments have an associated plunger which when depressed causes serrated cutters to puncture the foil seal and allow the contents to be added to the liquid containing or comprising the sample.

Methods for the diagnosis of visceral, cutaneous and canine leishmaniasis in a subject suspected of being infected with the parasitic protozoa Leishmania is disclosed. Disclosed are antibody-capture enzyme-linked immunosorbent assays (ELISAs) for the detection of antibodies to Leishmania parasite soluble antigens and antigen-capture ELISAs for the detection of Leishmania parasite soluble antigens in host samples. Also disclosed are immunodiagnostic kits for the detection of Leishmania parasite circulating antigens or IgM and IgG antibodies in a sample from subject having visceral, cutaneous or canine leishmaniasis. In these methods and kits, detection may be done photometrically or visually. The methods and kits also allow the visualization of Leishmania amastigotes or promastigotes in a sample.
