摘要: An apparatus for culturing cells includes a bioreactor. The bioreactor may be modular and may include in a chamber a fixed bed, such as an unstructured or structured fixed bed (such as a spiral bed) for culturing cells, with a return column arranged centrally within the chamber. The modular bioreactor may include a plurality of structured fixed bed arranged in a stacked configuration. The modular bioreactor may include an outer casing forming a space for conditioning (e.g., insulating, heating, cooling) at least a chamber in which cells are cultured. The bioreactor may also include an impeller with radially curved blades, and may also suspend the impeller so that it may move from side-to-side and align with an external drive. Related methods are also disclosed.

摘要: A packed-bed bioreactor is provided that includes: a vessel having an interior cavity defined by an outer wall; and a center column disposed within the interior cavity. The center column includes a columnar sidewall defining an inner region within the center column, the columnar sidewall separating the inner region from an outer region within the interior cavity. The bioreactor further includes a cell culture substrate disposed in the outer region of the cavity, the cell culture substrate surrounding the center column; at least one port extending through the vessel for at least one supply and removal of media to or from the interior cavity; and a fountain head element disposed above the center column.

摘要: We have found a way to make possible large-scale plasmid transfection using PEI to produce high titer viral vectors in fixed bed or adherent cell culture bioreactors by using PEI as a transfection agent, while avoiding formation of the PEI-plasmid precipitate which in prior art approaches clogged adherent bioreactor substrates. We have also found a way to improve PEI-based transfection by modifying how pH and CO2 are managed during transfection.

摘要: The present disclosure relates, according to some embodiments, to systems and methods for processing organic compounds, such as manure or other organic waste. Embodiments may comprise a first containment chamber, a first anaerobic chamber, and a second anaerobic chamber. A first anaerobic chamber may receive organic compounds from a first containment chamber, and may provide a fluid stream to a second anaerobic chamber. The second anaerobic chamber may comprise a substrate, such as lava rock, with bacteria growing thereon. Further, a sulfide gas treating unit may receive and treat sulfide gases from a first anaerobic chamber and/or a second anaerobic chamber. A water storage unit may receive and store waste water or effluent from a first anaerobic chamber and/or a second anaerobic chamber.

When a cell detaching step at least comprising shaking or spinning the culture chamber is performed, the unoccupied space is large enough for the porous matrices collide to each other or to the culture chamber to generate momentum whereby the cells detach from the porous matrices to harvest the cells.
