Tangential flow filtration device is provided wherein liners are provided between the filtration element and the top and bottom holders or manifolds. The liners incorporate the flow channels and inlet and outlet ports, as well as a sensor mount. The liners are made of an inexpensive material and therefore are disposable after a single use, making it more cost effective to dispose of them than to clean the conventional manifolds. The sensor mount accommodates a removable sensor, and isolates it from the fluid path.
Abstract:
A method of filtering a liquid feed is described, comprising passing a liquid feed through a single pass tangential flow filtration (SPTFF) system and recovering the retentate and permeate from the system in separate containers. A method of filtering a liquid feed is also described comprising passing a liquid feed through a tangential flow filtration (TFF) system, recovering permeate and a portion of the retentate from the system in separate containers without recirculation through the TFF system, and recirculating the remainder of the retentate through the TFF system at least once. The methods of the invention can be performed using an SPTFF or a TFF system that comprises manifold segments to serialize the flow path of the feed and retentate without requiring diverter plates.

A method and device for packing a chromatography column formed of one or more vibration devices attached to top and/or bottom flanges of the column. Media is added in one or more steps to the column, allowed to settle under the effects of gravity and then subjected to one or more treatments of vibration from the vibration devices until a suitably packed column is obtained. Liquid used to suspend the media while being placed into the column may be at least partially removed before or during the vibration step(s). The remaining liquid is then removed or replaced after the packing has been obtained.

A device for supporting a plurality of flexible containers for liquid, including a plurality of baskets, each containing at least one of the flexible containers, a magazine to receive the baskets, able to adopt an operating configuration in which the baskets are disposed inclined one above the other, which magazine includes supports for the baskets, able to adopt a loading/unloading position such that each basket can be loaded/unloaded along a horizontal path, and able to adopt an operating position to maintain each basket in an inclined position to make the magazine adopt the operating configuration, and a driver for driving the supports between the loading/unloading position and the operating position.

Fluid transfer device and method that allows for sterile wet connections, allows the connection to be reversed, and allows the connection to be reconnected, while leaving the connectors sterile and reusable under pressure. In certain embodiments, the device includes a first member and a second member, the latter adapted to receive the former in locking engagement upon actuation of the device to create fluid communication between the two in a sterile manner. Each of the members include a door that when opened, allows the first member to be linearly displaced into the second member to allow fluid to be transferred. When fluid transfer is complete, the first member can be retracted from the second member, and the doors closed.

In other embodiments, the invention relates to a process for increasing the capacity of a chromatography device for a target protein by delivering a concentrated sample comprising the target protein to a chromatography device.

Abstract:
A porous electrospun polymeric nanofiber liquid filtration medium, such as an electrospun mats, used for the removal of viral particles (e.g., parvovirus) and other particles in the 18 nm to 30 nm size range from fluid streams, having a mean flow bubble point measured with perfluorohexane above 100 psi. The electrospun medium includes nanofibers having an average fiber diameter of about 6 nm to about 13 nm, and the nanofiber liquid filtration medium has a mean pore size ranging from about 0.01 um to about 0.03 um, a porosity ranging from about 80% to about 95%, a thickness ranging from about 1 um to about 100 um, and a liquid permeability greater than about 10 LMH/psi. The high porosity of the electro-spun mats enable much higher water fluxes, thus reducing the time required to complete virus filtration steps on a fluid stream.
