A preparation method of drug-loaded micelles of an absorbable vascular stent coating for angiostenosis in an infant is provided, including the following steps: S1: dissolving a drug to be encapsulated in an appropriate amount of an emulsifying agent, adding a chitosan-poly(p-dioxohone) amphiphilic block copolymer (chitosan-b-PPDO copolymer), and thoroughly mixing to obtain a drug-copolymer solution; S2: adding the drug-copolymer solution obtained in S1 dropwise to an emulsifying agent aqueous solution prepared in advance, and continuously stirring to obtain a stable drug-loaded micellar solution; and S3: removing the emulsifying agent from the micellar solution obtained in S2 through vacuum evaporation, stirring a resulting concentrate, and centrifuging the concentrate to obtain a supernatant; and filtering the supernatant to obtain a filtrate, and subjecting the filtrate to dialysis to obtain the drug-loaded micelles.

The present invention provides a novel biomaterial which is a hybrid, self-assembling biopolymeric networked film that is functionalized through hydrophobic interactions with vesicles loaded with bioactive agents. The biomaterial compound is a polymeric network of hydrophobically modified chitosan scaffolds that is taken from solution and formed as a solid film. This solid state film is capable of hydrophobic interactions with the functionalized vesicles. The vesicles include one or more lamellar structures forming one or more nano-compartments that are capable of containing similar or alternative active moieties within. Use of the film results in a degradation of the chitosan scaffold thereby releasing the active moieties within the vesicles from the scaffold. Application of the current invention occurs through various delivery mechanisms and routes of administration as will be described herein.

Devices and methods for balloon delivery of rapamycin and other hydrophobic compounds to the wall of blood vessels. Balloon catheters, such as those used for balloon angioplasty, are modified with the addition of a reservoir of dry micelles. The micelle preparation is reconstituted and the micelles are mobilized when the aqueous solution used to inflate the balloons is injected into the catheter. The micelles are infused into tissue surrounding the balloon when pressurized fluid within the balloon leaks through the wall of the balloon.
