Abstract: Disclosure relates to a device and a method of using the device for conducting biological and chemical assays on a sample. The device includes two plates. The first plate includes at least a first and a second sample contact areas on its inner surface. Each sample contact area includes spacers having a bottom end fixed on the inner surface and a top end that is distal to the inner surface. The spacers in the first sample contact area differ in height from those in the second sample contact area. The first and second sample contact areas are disposed at different depths along the inner surface of the first plate, so that the top ends of the spacers are aligned on the same flat plane. The method includes depositing the sample onto the sample contact areas, compressing the sample into a thin layer, and analyzing the sample.

Abstract: A device and method are proved for transferring a target from a first location to a second location. The target is bound to solid phase substrate to form a target bound solid phase substrate. The device includes transfer surface for receiving the target bound solid phase substrate thereon for transfer. The transfer surface movable between a first position wherein the transfer surface is aligned with the first location and spaced therefrom by a distance and a second position wherein the transfer surface is aligned with the second location. An alignment structure aligns the transfer surface with respect to the second location, with the transfer surface in the second position. A force is movable between an attraction position wherein the target bound solid phase substrate are drawn toward the transfer surface and a discharge position wherein the target bound solid phase substrate are free of the force.

Abstract: In accordance with embodiments herein a method for capturing cells of interest in a digital microfluidic system is provided, comprising utilizing a droplet actuator to transport a sample droplet to a microwell device. The microwell device includes a substrate having a plurality of microwells that open onto a droplet operations surface of the microwell device. The sample droplet includes cells of interest that enter the microwells. The method introduces capture beads to the microwells, and the capture elements are immobilized on the capture beads. The method utilizes the droplet actuator to transport a cell lysis reagent droplet to the microwell device. Portions of the cell lysis reagent droplet enter the microwells and, during an incubation period, cause the cells of interest to release analyte that is captured by the capture elements on the capture beads.

Abstract: A micro-structured substrate to support a liquid sample having a lower face and an upper face. At least one surface cavity of volume V0 opening onto the upper face and forming a zone for analyzing the liquid sample. A first groove having a first volume V1, positioned around each surface cavity and opening onto the upper face thereof. The substrate also includes at least one second groove opening onto the upper face thereof and positioned around the first groove. The sum of the volumes of the second grooves is equal to V2. The volume V1+V2 is greater than or equal to 0.05 V0, preferably 0.1 V0. An analysis assembly including the substrate and the use thereof is also contemplated herein.
