This method eliminates the necessity of using whole chicken embryos for the production of Influenza vaccines. The method also provides for the continuous or batch-wise removal of culture media. As such, the present invention allows the large-scale continuous production of viruses to a high titer.

The present invention provides methods of reducing the levels of a titratable selectable pressure required, the number of amplification cycles, and the time taken to generate protein expressing cell lines by altering the codons of the desired open-reading-frames. Through the use of codon adaptation for this purpose the methods of the invention consistently provide sufficient yields in faster time frames saving many weeks in cell line development activities. Furthermore the methods of the invention also generate cell lines with lower concentrations of selection and amplification agent than previously achievable. Accordingly lower levels of selection and amplification marker in the final cells lines are observed.

The invention relates to a biomass for producing virus/virus antigen, which consists of cell aggregates of avian embryo cells having diameters of between 100 .mu.m and 1,000 .mu.m. The biomass according to the invention has a high metabolic activity in suspension in the culture medium and is infected with virus. It enables the large-scale production of pure virus/virus antigen and is particularly suitable for the production of TBE-virus/virus antigen.
