摘要: An image flow cytometer for observing a microparticulate sample includes a flow chamber having a flow channel that permits the microparticulate sample to travel in a flow direction. An irradiation system scans an irradiation spot across a sensing area of the flow channel in a scan direction different from the flow direction. A detection system detects resultant light from the sensing area and provides a detection signal. An alignment system alters a location of the sensing area with respect to the flow chamber. A control unit causes the irradiation system to scan the irradiation spot during a first measurement interval and operates the alignment system to translate the location of the sensing area along the flow direction. The flow chamber can be mounted to a movable stage in some examples, and the alignment system can move the flow chamber substantially opposite the flow direction using the stage.

The second data processing part generates, as indication values for gain adjustment of the light-receiving signal, the second indication value showing the intensity of a light-receiving signal obtained from the particle analysis apparatus, which is different from the aforementioned first indication value. The indication value output part outputs the first and second indication values to a display device.

摘要: Aspects and embodiments of the instant disclosure provide a particle and/or intracellular organelle alignment agent for a particle analyzer used to analyze particles contained in a sample. An exemplary particle and/or intracellular organelle alignment agent includes an aqueous solution, a viscosity modifier, and/or a buffer. Embodiments also encompass systems, compositions, and methods for analyzing a sample containing particles. Parrticles such as blood cells can be categorized and counted by a digital image processor. A digital microscope camera can be directed, for example using certain focusing techniques, into a flowcell defining a symmetrically narrowing flowpath in which the sample stream flows in a ribbon flattened by flow and viscosity parameters between layers of sheath fluid. Blood cell images can be collected and analyzed using dynamic range extension processes and systems.

摘要: The present disclosure relates to apparatus, systems, compositions, and methods for analyzing a sample containing particles. A particle imaging system or analyzer can include a flowcell through which a urine sample containing particles is caused to flow, and a high optical resolution imaging device which captures images for image analysis. A contrast pattern for autofocusing is provided on the flowcell. The image processor assesses focus accuracy from pixel data contrast. A positioning motor moves the microscope and/or flowcell along the optical axis for autofocusing on the contrast pattern target. The processor then displaces microscope and flowcell by a known distance between the contrast pattern and the sample stream, thus focusing on the sample stream. Cell or particle images are collected from that position until autofocus is reinitiated, periodically, by input signal, or when detecting temperature changes or focus inaccuracy in the image data.

摘要: A method for generating a three-dimensional luminescence image includes setting a focal interval between two-dimensional images in accordance with localization of luminescence in a three-dimensional sample. The three-dimensional sample contains a plurality of cells prepared to be luminescent and has a three-dimensional shape. The two-dimensional images have mutually different focal planes and are acquired at the focal interval. The method further includes acquiring a two-dimensional image set including two-dimensional images at the focal interval that is set by imaging the three-dimensional sample under an unirradiated condition; and generating a three-dimensional luminescence image by combining the two-dimensional images included in the two-dimensional image set together.

摘要: Platelets or blood cells are detected in a fluid sample by adjusting a focal depth of a microscope through a range of values, the microscope having a mounted sample and an objective lens adapted with one or both of (a) a spherical aberration correction unmatched to a utilized cover plate for the sample, or (2) a numerical aperture unmatched to a utilized illumination source for the sample. Images are recorded at different specific focal depths and in multiple z planes of a fluid bearing the platelets, where the position of platelets may overlap on different of the multiple z planes that are recorded, the images recorded through the cover plate, thus causing the generation of a specific light-dark pattern indicative of platelets at particular positions and at multiple depths in the fluid media. The images are analyzed for the specific light-dark pattern.

摘要: Flow cytometer systems are provided having intermediate angle scatter detection capability. In some aspects, systems are provided that include an intermediate angle scatter (IAS) light detector positioned to measure intermediate angle scatter emitted from a flow cytometer. The system further includes a mask disposed across a portion of the IAS light detector and positioned between the flow cell and the IAS light detector to cover at least a central portion of the IAS light detector so as to block a diffraction pattern observed at the detector. In some instances, the diffraction pattern is created by a flat beam profile irradiating the sample. Methods are also provided for configuring a flow cytometer to block a diffraction pattern created by (1) a flat laser beam profile irradiating a flow cytometer liquid sample, or (2) a mismatched index of refraction between a sheath fluid and a liquid sample in a flow cytometer.

摘要: A method for detecting bacteria and determining the concentration thereof in a liquid sample includes the steps of taking an optical section through a container holding a volume of the liquid sample at a predetermined field of view and at a predetermined focal plane depth or angle and after a period of time has elapsed to allow non-bacteria in the sample to settle to the bottom of the container. Since bacteria auto arranges in the liquid sample, forming a lattice-like grid pattern, an optical section through the volume of auto-arranged bacteria may be used to measure the quantity of bacteria residing in that section. A container for holding the liquid sample has particular structure which aids in separating the non-bacteria from the bacteria.
