    摘要: IT HAS BEEN FOUND THAT VERY LARGE PRODUCTION OF ANTIBODIES CAN BE ACHIEVED BY REMOVING SPECIFIC FEEDBACK REGULATORY ANTIBODIES BY MEANS OF A LYMPHORESIS PERFORMED UNDER SPECIAL CONDITIONS IN A PATIENT OR SUBJECT (E.G., AN ANIMAL OR HUMAN) WITH INDUCED ANATOMICAL AND PHYSIOLOGICAL CHANGES. THE SUBJECT IS FIRST GIVEN ANTIGEN ADMINISTRATION THEN, PREFERABLY, BUT NOT MANDATORILY, IS SPLENECTOMIZED. A THORACIC DUCT FISTULA IS NEXT PERFORMED. THE CENTRAL VENOUS SYSTEM PRESSURE IS THE PREFERABLY RAISED SO THAT IT IS ABOVE THE ATMOSPHERIC PRESSURE OF THE THORACIC DUCT. IN THIS MANNER, SUBSTANTIALLY ALL THE LYMPTH FLUID IS ALLOWED TO FLOW OUT OF THE THORACIC DUCT FROM THE FIISTULA (THROUGH AN INDWELLING CATHETER) FOR A PROLONGED PERIOD OF TIME. THE LYMPH IS SEPARATED INTO CELLS AND LYMPH FLUID. THE CELLS ARE RETURNED TO THE SUBJECT INTRAVENOUSLY. THE SUBJECT MUST BE GIVEN REPLACMENT FLUID, WHICH CAN BE OF SEVERAL KINDS, BUT ALL LACKING THE SPECIFIC ANTIBODY. BY VIRTUE OF THE ABOVE PROCEDURE, THE PLASMA AND EXTRACELLULAR FLUID OF THE SUBJECT IS CONTINUOUSLY DEPLETED OF FEEDBACK ANTIBODY BECAUSE THE ANTIBODY IS CONTINUOUSLY REMOVED PRIOR TO ITS REACHING THE BLOOD STREAM. BECAUSE OF THIS LACK OF ANTIBODY, IN THE PRESENCE OF ANTIGEN ADMINISTRATION, IT IS FOUND THAT THE ANTIBODY PRODUCTION IN THE LYMPHOID TISSUE AND THEREFORE ITS CONTENT IN THE LYMPH FLUID IS ENORMOUS AND EVER-INCREASES. THE TREMENDOUS INCREASE IN ANTIBODY PRODUCTION APPEARS TO BE SEVERAL ORDERS OF MAGNITUDE GREATER THAN OTHER MODES OF ANTIBODY PRODUCTION AND THEREFORE APPEARS TO HAVE VERY SUBSTANTIAL UTILITY IN THE FIELDS OF BIOLOGY, CHEMISTRY AND VETERINARY AND CLINICAL MEDICINE.

    摘要: This invention provides a complex consisting essentially of prealbumin, retinol-binding protein, and a retinoid which binds to retinol-binding protein, wherein the concentration of retinoid in the complex is from about 2.times.10.sup.-5 M to about 10.sup.-10 M and the concentration of prealbumin and retinol-binding protein is sufficient to maintain the retinoid concentration from about 2.times.10.sup.-5 M to about 10.sup.-10 M. This invention also provides a composition for enhancing the growth of human B cells in culture which comprises an amount of the complex of this invention effective to enhance the growth of human B cells and a nutrient medium. This invention further provides a method of enhancing growth of human B cells in culture comprising growing the cells in culture in a growth medium containing a concentration of the complex effective to enhance growth of the cells. This invention also provides a synthetic growth medium for culturing animal cells comprising the complex of this invention at a concentration effective to enhance cell growth. This invention also provides a method of preventing the growth of human tumor cells containing growth factor which comprises removing the growth factor from the human tumor cells.

    摘要: What are disclosed are an isolated tissue protein, denominated PP16, which can be used to prepare anti-sera useful as diagnostic agents, said protein being found in extracts of some human organs such as the placenta, spleen, stomach, and lung and being capable of isolation therefrom and which has the following properties:(a) a carbohydrate content of 4.3.+-.1.6%, comprising 3.4.+-.1.2% of hexoses, 0.24.+-.0.07% hexosamines, 0.06.+-.0.03% of fucose, and 0.6.+-.0.3% of neuraminic acid;(b) a sedimentation coefficient S.sub.20,w.sup.0 of 4.6.+-.0.4 S;(c) a molecular weight, determined in a polyacrylamide gel containing sodium dodecyl-sulfate, of 46,000.+-.3,000;(d) an extinction coefficient E.sub.1 cm.sup.1% (280 nm) of 8.82.+-.0.5;(e) an electrophoretic mobility in the range of albumin; and(f) an isoelectric point of 4.7.+-.0.2.
