摘要: An example image sensor structure includes an image layer. The image layer includes an array of light detectors disposed therein. A device stack is disposed over the image layer. An array of light guides is disposed in the device stack. Each light guide is associated with at least one light detector of the array of light detectors. A passivation stack is disposed over the device stack. The passivation stack includes a bottom surface in direct contact with a top surface of the light guides. An array of nanowells is disposed in a top layer of the passivation stack. Each nanowell is associated with a light guide of the array of light guides. A crosstalk blocking metal structure is disposed in the passivation stack. The crosstalk blocking metal structure reduces crosstalk within the passivation stack.

摘要: Methods are provided for separating magnetically responsive beads from a droplet in a droplet actuator. Droplet operations electrodes and a magnet are arranged in a droplet actuator to manipulate a bead-containing droplet and position it relative to a magnetic field region that attracts the magnetically responsive beads. The droplet operations electrodes are operated to control the droplet shape and transport it away from the magnetic field region to form a concentration of beads in the droplet. The continued transport of the droplet away from the magnetic field causes the concentration of beads to break away from the droplet to yield a small, concentrated bead-containing droplet immobilized by the magnet.

摘要: A stack of fluidics layers of a microfluidic cartridge for sequencing nucleic acid molecules includes a sequencing chamber layer having a sequencing chamber area configured for carrying out clustering and sequencing reactions, and a sequencing chamber bottom layer disposed under the sequencing chamber layer. The sequencing chamber bottom layer has an opening configured to hold an image sensor with the image sensor having an active area disposed under the sequencing chamber area. The sequencing chamber area spans substantially all of the active area of the image sensor. The stack of fluidics layers includes a flexible printed circuit board (PCB) layer under the sequencing chamber bottom layer, and a fluidics channels layer disposed under the flexible PCB layer. The fluidics channels layer includes fluidics channels that are configured to deliver reactants to the sequencing chamber area. The fluidics channels do not substantially overlap with the active area of the image sensor.

摘要: Techniques are described for dynamically correcting image distortion during imaging of a patterned sample having repeating spots. Different sets of image distortion correction coefficients may be calculated for different regions of a sample during a first imaging cycle of a multicycle imaging run and subsequently applied in real time to image data generated during subsequent cycles. In one implementation, image distortion correction coefficients may be calculated for an image of a patterned sample having repeated spots by: estimating an affine transform of the image; sharpening the image; and iteratively searching for an optimal set of distortion correction coefficients for the sharpened image, where iteratively searching for the optimal set of distortion correction coefficients for the sharpened image includes calculating a mean chastity for spot locations in the image, and where the estimated affine transform is applied during each iteration of the search.

摘要: A droplet actuator device for conducting droplet operations is provided that comprises a substrate defines a device channel to conduct droplet operations. Electrodes are arranged proximate to the substrate. A drive circuit is connected to the electrodes. The drive circuit generates an electrode drive signal to drive the droplet operations based on a reference waveform. The electrode drive signal is partitioned into an AC modulated drive cycle formed of sub-cycles. The electrode drive signal switches, during the sub-cycle, between at least first and second states where a degree of modulation with respect to the reference waveform forms a balanced modulation pattern.
