摘要: Methods of making a spheroid are provided whereby a suspension is first produced including one or more biologically-relevant materials dispersed within a biocompatible medium. A droplet of the suspension is then bioprinted into a salt solution by bringing the droplet into contact with a surface of the salt solution in a controlled manner to reproducibly yield a spheroid containing a desired size and a desired amount of biologically-relevant materials.

摘要: Disclosed herein is a bioreactor system that allows perfusive flow through a porous support medium enabling 3D growth of biological samples. In some embodiments, the system comprises a sample well filled with a three-dimensional (3D) cell growth medium. The system can further comprises a liquid medium reservoir fluidly connected to the sample well by a first filter material. The system can further comprises a medium collection chamber fluidly connected to the sample well by a second filter material. The system can further comprise an absorbant material that creates an osmotic pressure gradient to produce perfusive flow. In some embodiments, osmotic pressure draws fluid from the liquid medium reservoir, through the first filter material, into the sample well where it permeates the three-dimensional cell growth medium, through the second filter material, and finally into the medium collection chamber.

摘要: Provided are a cell culture device for producing a cell sheet to be used in regenerative medicine and drug discovery research, and a kit for producing a cell sheet. Specifically, a cell detachment inhibition region having cell adhesive force stronger than that of a stimulus-responsive region regardless of the presence or absence of stimulation, and inhibiting cell detachment is directly provided over the entire circumference of a cell culture surface, and the stimulus-responsive region having adhesiveness to cells, which is varied due to an external stimulus, is arranged inside the cell detachment inhibition region, thereby forming a culture device for prevention of cell detachment due to mechanical stimulation such as medium exchange. Moreover, with a support for harvesting cell sheet to be used for the harvest of a cell sheet having given features, the cell sheet can be easily harvested.

摘要: The invention provides a device for growing cells—referred to as a cassette. The cell culturing device includes a housing that contains a lid having an optically clear window; a fluid distribution channel; a sample injection port fluidically connected to the fluid distribution channel; a base housing a porous media pad; and a media injection port fluidically connected to the media pad. The lid mates to the base to form a sterile seal; the fluid distribution channel is disposed over the media pad, which is viewable through the optical window; and sample fluid introduced into the fluid distribution channel is distributed evenly to the media pad, e.g., via a plurality of channels. The invention also provides kits that include cassettes of the invention and a tube set.

摘要: A method for detecting oncogenic growth and viability, and/or degree of cellular transformation and/or identifying an agent that inhibits cellular transformation is disclosed. The method including: providing a cellular sample, such as a sample of cells obtained from a subject or a cell line; culturing the cellular sample in low attachment conditions; and detecting growth and7or cell viability of the sample, wherein increased growth relative and/or viability relative to a control or control level indicative of basal growth and/or viability indicates cellular transformation. In some embodiments, the method includes introducing a n expression vector into cells of the cellular sample, wherein the expression vector comprises a gene product expression sequence being tested for transformation ability. In some embodiments the cellular sample is contacted with a test agent and growth and/or cell viability of the sample is determined to determine if the agent inhibits transformation.

A semipermeable membrane is disposed at a bottom of the cell retention vessel, and a frame comprising a grid is disposed underneath the semipermeable membrane.

The unit has an upright orientation and a reclined orientation in which the axis of rotation is at a reclined angle to horizontal relative to the upright orientation. The digestion drive unit is configured to receive and drive rotation of the tissue processing unit in the reclined orientation. Methods for processing biological material including adipose tissue include enzymatically digesting adipose tissue with rotation of a tissue processing unit around an axis of rotation in a reclined orientation.
