Abstract: A cell processing system includes a first processor connectable to a source container filled with a biological fluid, a second processor, and a controller coupled to the processors. The first processor includes a separator configured to separate the biological fluid into at least two streams of material, and a first container configured to receive one of the streams. The second processor includes a magnetic separator configured to select target cells, the target cells being associated with magnetic particles, a second, pass-through container associated with the magnetic separator, the second container connected at a first end to the first container, and a third container connected to a second end of the pass-through container. One of the processors includes at least one pump configured to transfer material between the separator and the first container, and between the first container and the second container.

Abstract: The invention herein relates to conducting assays with an apparatus including a substantially transparent assay cartridge loaded with magnetic beads, and a magnet carrier base positioned below a scanning platform holding the assay cartridge. The assay cartridge includes magnetic beads, sample and control solutions in some wells, and assay reagents in others. A microcomputer controls a stepping motor which controls movement of the magnet carrier base, and causes the magnetic beads to travel from one well to another. An electromagnetic coil-spring assembly induces mixing of well contents with the magnetic beads on actuation. The assay cartridge is authenticated by sending its encoded identifier to a server or website, and assay instructions are provided remotely to the microcomputer. Following assay completion, the cartridge can have color change or other assay indication detected, and the results sent to the server or website or another recipient.

Abstract: An assay device including a loading zone for a sample that may contain an analyte, an activator in communication with the loading zone from which at least one activator is displaced by presence of the analyte, an amplifier in an inactive state in communication with the activator that becomes activated in the presence of the activator, a biomatrix barrier in communication with the activated amplifier that is degraded or modified by the activated amplifier, and an indicator responsive to the degradation or modification of the biomatrix barrier, which in turn reflects a concentration of the analyte in the sample loaded on the device. The selection of analyte or displacement of the activator by presence of the analyte may occur off-platform and/or may rely on use of non-covalent interactions. Abstract: Methods of assaying a biological target are disclosed. The method comprises: (a) providing a sample containing the biological target; (b) providing biotin-labeled first aptamers conjugated to a gold nanoparticle (GNP), and second aptamers conjugated to a magnetic bead, wherein the first and the second aptamers exhibit specific binding affinities to the target; (c) incubating the sample with the first and the second aptamers to obtain target-bound aptamers; (d) separating the target-bound aptamers from unbound aptamers; (e) eluting the first aptamers from the GNP; (f) incubating the eluted biotin-labeled first aptamers with streptavidin-magnetic beads and reporter gold nanoparticles (GNPs) to obtain a complex comprising: the bead, the first aptamers, attached to the bead; and the reporter GNPs captured by the bead through the first aptamers; (g) eluting the reporter GNPs captured; and (h) detecting the target by measuring and analyzing a light-scattering signal of the eluted reporter GNPs.
