Abstract: Systems and methods for detecting and processing signals from particles. In an exemplary method, particles may be passed through a zone of a channel, while the zone is irradiated with light. Interaction of the light with the particles may deflect light and induce photoluminescence. A deflection signal and a photoluminescence signal may be detected from the zone. Particle waveforms may be identified in the deflection signal. At least a subset of the particle waveforms may be double-peak waveforms including a pair of peaks corresponding to a particle entering and exiting the zone. Amplitudes may be obtained from the photoluminescence signal. The amplitudes may correspond to respective particles and their particle waveforms, and at least a subset of the amplitudes may correspond to the double-peak waveforms. Individual particles may be assigned as positive or as negative for an analyte based on the corresponding amplitudes.

Abstract: A container assembly for use with a high-pressure liquid chromatography (HPLC) instrument is disclosed, in which the container assembly, when coupled to a source of pressurized gas, provides fluid medium to the HPLC instrument at positive pressure. The container assembly has an external exterior container shell, an internal fluid container for holding fluid medium, an interstitial volume between the external exterior container shell and the internal fluid container, a port for fluidly connecting the volume to a pressurized gas source, and a port for fluidly connecting the internal fluid container to the HPLC instrument. As a pressurized gas in the interstitial volume increases, fluid medium flows out of the port connected to the internal fluid bag and container assembly at a positive pressure. A system incorporating the container assembly, and method of use of the same, are also disclosed.

Abstract: Systems and methods for producing blot images. A blot, for example a western blot, is imaged using an imaging system having a field of view and a magnification. Features of interest in the blot correspond to features in the digital image, and the sizes of the features in the digital image depend on the magnification of the imaging system. A structuring element is selected based on the sizes and shapes of the features in the digital image, and the image is morphologically eroded and dilated varying numbers of times. The eroded and dilated image is subtracted from the original blot image to remove background signal from the blot image, producing an output image. The number of erosions needed to completely erode the features of interest is determined automatically, for example by investigating the behavior of the kurtosis of the output image as a function of the number of erosions performed.
