摘要: Embodiments herein relate to cell encapsulation membranes, devices including the same, and related methods. In an embodiment, a cell encapsulation membrane is included. The cell encapsulation membrane can include a mesh substrate. The mesh substrate can include a first series of fibers extending in a first direction and a second series of fibers extending in a second direction, the first series of fibers intersecting with the second series of fibers, the mesh substrate defining a plurality of apertures disposed between adjacent fibers of the first series and the second series. The cell encapsulation membrane can further include a coating disposed on the mesh substrate, the coating partially occluding the plurality of apertures defined by the mesh substrate and forming pores. Other embodiments are also included herein.

The biologic is generally adapted to controllably release the agent to the surrounding tissue or organ to provide a local and targeted delivery.

摘要: A composition containing biocompatible hydrogel encapsulating mammalian cells and anti-inflammatory drugs is disclosed. The encapsulated cells have reduced fibrotic overgrowth after implantation in a subject. The compositions contain a biocompatible hydrogel having encapsulated therein mammalian cells and anti-inflammatory drugs or polymeric particles loaded with anti-inflammatory drugs. The anti-inflammatory drugs are released from the composition after transplantation in an amount effective to inhibit fibrosis of the composition for at least ten days. Methods for identifying and selecting suitable anti-inflammatory drug-loaded particles to prevent fibrosis of encapsulated cells are also described. Methods of treating a disease in a subject are also disclosed that involve administering a therapeutically effective amount of the disclosed encapsulated cells to the subject.

摘要: Adult autologous stem cells cultured on a porous, three-dimensional tissue scaffold-implant for bone regeneration by the use of a hyaluronan and/or dexamethasone to accelerate bone healing alone or in combination with recombinant growth factors or transfected osteogenic genes. The scaffold-implant may be machined into a custom-shaped three-dimensional cell culture system for support of cell growth, reservoir for peptides, recombinant growth factors, cytokines and antineoplastic drugs in the presence of a hyaluronan and/or dexamethasone alone or in combination with growth factors or transfected osteogenic genes, to be assembled ex vivo in a tissue incubator for implantation into bone tissue.
