Abstract: A method for selectively recovering nucleic acid from a sperm cell in a sample containing cells of at least a sperm cell and an epithelial cell, and a cell suspension medium comprising extracellular impurities, is provided. The method entails introducing a sample into a vessel, sequestering the cells from the remaining sample components, washing the cells with a washing solution either before or after sequestration, removing the impurities-containing cell suspension medium from the vessel while retaining the cells; lysing selectively cells of the first cell type; and isolating the nucleic acid from the lysed cells. Methods for recovering nucleic acid from the second cell type are also provided.

Abstract: A method of preparing reagents includes inserting a cartridge into an instrument. The cartridge includes a plurality of reagent enclosures disposed in a cavity of the cartridge and exposing a port to an exterior of the cartridge. Each reagent enclosure includes a reagent container including a reagent and an internal cavity defining a compressible volume, an opening defined through the reagent container to the internal cavity. The method further includes connecting a plurality of fluid ports to the openings of the plurality of reagent enclosures; applying a solution through the fluid ports to at least partially fill the plurality of reagent enclosures; and cycling a pressure of the cavity, whereby for each of the reagent enclosures, during increasing pressure, the solution enters the internal cavity of the reagent container, combines with the reagent, and compresses the compressible volume, and during decreasing pressure, the compressible volume decreases and the reagent is ejected through the opening.

Abstract: A polymer substrate, such as a polymer particle, is formed from a carboxyl functional monomer. In an example, the carboxyl functional monomer has a protection group in place of the OH of the carboxyl group. Once the monomer is polymerized, such a protection group can be removed, providing a polymer network with carboxyl functional sites. Such sites can be used to attach other functionality to the polymer substrate.
