摘要: The presently claimed and described technology provides integrated automated instruments (100, 100A, 100B, 100C) and methods (800) for automatically processing a whole blood sample (50) in a single sample tube (80). These include: mixing the sample in the tube; analyzing a portion (50P) of the mixed sample (50M); centrifuging the sample in the tube thereby separating plasma (60); and analyzing a portion (60P) of the plasma. A pretreatment module (200) may mix, centrifuge, transport, and store the sample. The integrated instrument may include multiple detector components and does not require external sample handling.

摘要: A method of determining glucose values during continuous glucose monitoring (CGM) measurements includes providing a CGM device including a sensor, a memory, and a processor; applying a constant voltage potential to the sensor; measuring a primary current signal resulting from the constant voltage potential and storing the measured primary current signal in the memory; applying a probing potential modulation sequence to the sensor; measuring probing potential modulation current signals resulting from the probing potential modulation sequence and storing measured probing potential modulation current signals in the memory; determining an initial glucose concentration based on a conversion function value and the primary current signal; determining a connection function based on the primary current signal and a plurality of the probing potential modulation current signals; and determining a final glucose concentration based on the initial glucose concentration and the connection function value. Other aspects are provided.

摘要: A blood sample collector can be used to collect a blood sample from a subject. The blood sample collector can be placed in a receptacle of a spectrometer to measure spectral data from the blood sample while the blood sample separates. The container may comprise a window to allow light such as infrared light to pass through the container, with the blood sample at least partially separating within the container between spectral measurements, which can provide improved accuracy of the measurements and additional information regarding the sample. The container may comprise an elongate axis and the container configured for placement in the spectrometer receptacle with the elongate axis extending toward a vertical direction in order to improve gravimetric separation of the blood sample. The spectrometer can be configured to measure the blood sample at a plurality of heights along the sample as the sample separates.

摘要: In one aspect, a method to detect white blood cells and/or white blood cell subtypes from non-invasive capillary videos is featured. The method includes acquiring a first plurality of images of a region of interest including one or more capillaries of a predetermined area of a human subject from non-invasive capillary videos captured with an optical device, processing the first plurality of images to determine one or more optical absorption gaps located in said capillary, and annotating the first plurality of images with an indication of any optical absorption gap detected in the first plurality of images. The method also includes acquiring a second plurality of images of the same region of interest of the same capillary with an advanced optical device capable of resolving cellular structure of white blood cells and white blood cell subtypes and spatiotemporally annotating the second plurality of images with an indication of any white blood cell detected and/or a subtype of any white blood cell detected in the second plurality of images. The method also includes inputting the first plurality of images and annotated information from the first plurality of images and annotated information from the spatiotemporally annotated second plurality of images into a machine learning subsystem configured to determine a presence of white blood cells and/or the subtype of any white blood cells present in the one or more optical absorption gaps in the first plurality of images.

The method may include comparing the first value to a first reference range. The method may also include comparing the second value to a second reference range. Furthermore, the method may include classifying an operational status of the hematology analyzer based on the comparison of the first value to the first reference range and based on the comparison of the second value to the second reference range.

摘要: A microfluidic, chip-based assay device has been developed for measuring physical properties of an analyte (particularly, whole blood or whole blood derivatives). The technologies can be applied to measure clotting times of whole blood or blood derivatives, determine the effects of anticoagulant drugs on the kinetics of clotting/coagulation, as well as evaluate the effect of anticoagulant reversal agents. These technologies can additionally be used to optimize the dosage of anticoagulation drugs and/or their reversal agents. The assay is independent of the presence of anticoagulant; clotting is activated by exposure of the blood sample in the device to a glass (or other negatively charged material such as oxidized silicon) surface, which activates the intrinsic pathway and can be further hastened by the application of shear flow across the activating materials surface. The absence of chemical activating agents and highly controlled and reproducible micro-environment yields a point of care universal clotting assay.
