In one embodiment, the apparatus includes a substrate having a set of electrical leads for connecting electrodes to a controller, a first set of electrodes, each connected to a separate one of the electrical leads, and a second set of electrodes, all connected to a single one of the electrical leads. In another embodiment, the apparatus includes a substrate having a set of X electrodes, and a set of Y electrical leads, each connected to one or more electrodes, wherein X is greater than Y.

摘要： Apparatuses for mixing droplets, such as a binary mixing apparatus, are provided. The binary mixing apparatus includes an array of electrodes and a conducting element positioned in relation to at least one of the electrodes to enable a droplet placed in electrical communication with the at least one electrode to electrically communicate with the conducting element. The binary mixing apparatus additionally includes an electrode selector for sequentially biasing one or more selected electrodes of the array to move a droplet disposed on the array into contact with another droplet. The apparatus further includes a first droplet supply area communicating with the array and a second droplet supply area communicating with the array.

摘要： In a method for sampling a continuous liquid flow, the liquid flow is supplied to a surface along an input flow path. The liquid flow is sampled by forming a sample droplet from a portion of the liquid flow. The sample droplet is moved along an analysis flow path to a processing area of the surface, where the sample droplet is processed. Discrete sample droplets are formed and moved using an electrowetting technique. A binary mixing apparatus and method are also provided. The apparatus comprises an array of electrodes, an electronic controller communicating with the electrodes, a sample droplet supply area communicating with the array, and an additive droplet supply area communicating with the array. The electronic controller alternately energizes and de-energizes selected electrodes to carry out droplet-to-droplet binary mixing operations to obtain one or more droplets having a target mixing ratio.

摘要： The present invention relates to droplet-based pyrosequencing including a method of identifying a base at a target position in a sample nucleic acid. The method includes: (a) providing a droplet microactuator including a first droplet including a sample nucleic acid immobilized on a bead; and (b) on the droplet microactuator: (i) contacting the first droplet with one or more reagent droplets to yield a second droplet, wherein the one or more reagent droplets include reagents for extending a double stranded portion of the sample nucleic acid by incorporating a nucleotide at the target position; (ii) splitting the second droplet to yield a third droplet including the bead and a fourth droplet lacking the bead; and (iii) assaying the third droplet to determine whether the nucleotide was incorporated at the target position.

摘要： The present invention relates to bead incubating and washing on a droplet actuator. Methods for incubating magnetically responsive beads that are labeled with primary antibody, a sample (i.e., analyte), and secondary reporter antibodies on a magnet, on and off a magnet, and completely off a magnet are provided. Also provided are methods for washing magnetically responsive beads using shape-assisted merging of droplets. Also provided are methods for shape-mediated splitting, transporting, and dispensing of a sample droplet that contains magnetically responsive beads. The apparatuses and methods of the invention provide for rapid time to result and optimum detection of an analyte in an immunoassay.
