    摘要: Cell based assays are performed in a microfluidic device, where the cells are introduced into a reservoir and are contacted with one or more agents prior to or during their residence in the reservoir or in a capillary channel connected to the reservoir. The cells are moved by electrokinesis individually from the reservoir to a detector, where the status of the cells as a result of contacting said agents is determined. Conditions are provided for moving the cells electrophoretically or by electroosmotic force, where the cells may be viable or fixed, natural or genetically modified.

    摘要: A dilution and mixing cartridge is provided that allows single (or multiple) dilutions of a sample with a diluent in a disposable cartridge in which a measurement, such as optical density, is made. Addition of sample to the device automatically measures the sample, and addition of diluent automatically causes a fixed ratio of sample and diluent to enter a receiving chamber, in which mixing and measurement can take place. A number of preferred embodiments are provided in which the diluent application site is located at a vertical height below the sample application site, flow directing chambers are provided to control flow of desired volumes of sample or diluted sample at the appropriate locations in the device and to direct excess sample and diluent to waste locations, or control mechanism are provided at flow directing chambers and other internal locations to verify correct operation of the device.

    摘要: Assay methods and compositions are provided for determining an analyte in a sample suspected of containing the analyte. The composition comprises in a novel single liquid reagent at least one specific binding pair (sbp) member and its complementary member wherein at least one sbp member is reversibly confined in a material that temporarily renders the confined sbp member incapable of binding with its complementary sbp member. At least one of the sbp members is bound to a member of a signal producing system capable of producing a detectable signal in relation to the amount of analyte in the sample. The confinement is reversed, any remaining members of the signal producing system are added, and the signal produced in relation to the amount of analyte is measured. Examples of the confining material are lipid bilayers, cells and gels.
