Abstract: Provided herein are compositions, systems, methods, and kits for joining together the ends of one or more polynucleotides using at least one pair of blocking oligonucleotide adaptors. Blocking oligonucleotide adaptors can comprise a double-stranded oligonucleotide adaptor (duplex) having an overhang cohesive portion that anneals with a blocking oligonucleotide which can be a separate single-stranded oligonucleotide. Also provided herein are compositions generated using the blocking oligonucleotide adapters, including circularized polynucleotides, that may be manipulated, for example, through nick translation, to yield a linear mate pair construct.

Abstract: A biological analysis system is provided. The system comprises an interchangeable assembly configured to accommodate any one of a plurality of sample holders, each respective sample holder configured to receive a plurality of samples. The system also includes a control system configured to cycle the plurality of samples through a series of temperatures. The system further includes an optical system configured to detect fluorescent signals emitted from the plurality of samples. The optical system, in particular, can comprise a single field lens, an excitation source, an optical sensor, and a plurality of filter components. The excitation source can be one or more light emitting diodes. The field lends can be a bi-convex lens.

The method includes calculating a total score for the candidate text phrase based on relative tag scores associated with each vocabulary tag for the one or more terms. The method includes classifying the candidate text phrase as a biological product reference and includes searching a database for one or more product entries based on the biological product references.

Abstract: The invention provides a passive fluidics circuit for directing different fluids to a common volume, such as a reaction chamber or flow cell, without intermixing or cross contamination. The direction and rate of flow through junctions, nodes and passages of the fluidics circuit are controlled by the states of upstream valves (e.g. opened or closed), differential fluid pressures at circuit inlets or upstream reservoirs, flow path resistances, and the like. Free diffusion or leakage of fluids from unselected inlets into the common outlet or other inlets at junctions or nodes is prevented by the flow of the selected inlet fluid, a portion of which sweeps by the inlets of unselected fluids and exits the fluidics circuit by waste ports, thereby creating a barrier against undesired intermixing with the outlet flow through leakage or diffusion. The invention is particularly advantageous in apparatus for performing sensitive multistep reactions, such as pH-based DNA sequencing reactions.

Abstract: Various embodiments of a low-volume sequencing system are provided herein. The system can include a low-volume flowcell having at least one reaction chamber of a defined volume (e.g., less than about 100 μl). The system can also include an automated reagent delivery mechanism configured to reversibly couple with the inlet port corresponding to a target reaction chamber thereby placing allowing for reagent to be accurately moved from a storage container to the reaction chamber with minimal reagent waste. The flowcells can include a plurality of reaction chambers (e.g., 6) thereby allowing for parallel analysis of multiple samples. Various methods of analyzing a biomolecule are also provided herein.

Abstract: An apparatus for preparation of an electrophoresis slab gel may include a base having an opening configured to receive a cassette configured to contain an electrophoresis slab gel, a clamping mechanism configured to move relative to the base between an open position in which the clamping mechanism permits insertion of a cassette into the base, and a closed position in which the clamping mechanism is configured to clamp a cassette received in the base, a compressible pad operatively coupled to the clamping mechanism in a position to compress against a cassette received in the base in the closed position of the clamping mechanism. The cassette may include a first plate and a second plate, and a spacer mechanism separate from each of the first and second plates, the spacer mechanism configured to be positioned between the inner faces of the first plate and the second plate. When subjected to a clamping force exerted on the outer faces of the first and second plates, the spacer mechanism is configured to maintain a separation distance between the inner faces of the first and second plates, and provide a seal to prevent leakage of an electrophoresis gel solution introduced between the plates.
