The fluidic circuit is also coupled to a plurality of chromatography columns. An interface to an analyzer receives output from at least one of the plurality of chromatography columns. The plurality of chromatography columns is moved relative to the analyzer via a translation stage, such that sample output from one of the plurality of chromatography columns can be selectively presented to the analyzer.

The present invention provides a method for the acoustic ejection of fluid droplets from fluid-containing reservoirs to form arrays suitable for high-throughput combinatorial crystallization experiments. Such arrays may utilize very small fluid volumes, in the order of picoliters. The method is especially suited to preparing combinatorial libraries useful in developing techniques for crystallizing biomacromolecules, such as proteins. The small volumes conserve macromolecules that may be costly and rare, and permit the testing of a large number of experimental crystallization conditions for a given amount of a macromolecule. The time required for the experiments may be very short due to the small volumes. The invention is conducive to forming high-density microarrays of small volume crystallization experiments. Acoustic detection of crystals in situ, and distinction between biomacromolecular and non-biomacromolecular crystals, are also taught.

A polymer synthesizer is disclosed which has a base on which sits a synthesis case, a synthesis block, and a means of moving the synthesis block and supports for a reagent shelf. The synthesis case has a loading station, drain station, and a water tolerant reagent area and a water sensitive reagent dispensing area. The synthesis case has a cover, a first and a second side, a first and a second end, and a bottom side which contacts the base. The bottom side of the synthesis case has a top face in which there are tracks. A synthesis block moves back and forth in the synthesis case and has a top face and an opening in the top face for a synthesis plate with a plurality of wells. The synthesis block also has a collection area under the synthesis plate to drain spent reagents or optionally hold a sample plate. The polymer synthesizer also has a means of moving the synthesis block back and forth in the synthesis case, preferably a pulley, cable and motor. The polymer synthesizer also has a means of draining the liquid from the synthesis plate. A method of synthesizing oligonucleotides with the polymer synthesizer also is disclosed.

A multi-channel fluid dispenser includes a reservoir with multiple internal chambers, a multi-channel liquid dispensing head, a plurality of fluid-delivery conduits, and support and positioning elements. The fluid-delivery conduits receive liquid from the chambers in the reservoir and deliver it to the dispensing head. The support and positioning elements are operable to support and position the dispensing head so that it can deliver the liquid from the reservoir to an underlying receiver. The fluid-delivery conduits include one or more optional flow control features that improve the accuracy of the dispensing operation. The multi-channel liquid dispensing head, which includes a valve-support member, a plurality of valves and a plurality of nozzles, is advantageously movable in three directions to facilitate alignment with an underlying receiver. Movement of the dispensing head along two axes is provided by the simple expedient of elongate holes having their long axes aligned in mutually orthogonal directions. In some embodiments, the valves are tiltable away from the vertical so that liquid dispensed from such tilted valves is directed at a non-normal angle into a receiver.

An apparatus for actuating a droplet comprises a first conductive layer, a second conductive layer, a conductive elongate element, and a voltage source. The first conductive layer comprises a first hydrophobic surface. The second conductive layer comprises a hydrophilic surface facing the first hydrophobic surface. The second conductive layer is axially spaced from the first conductive layer to define a gap therebetween. The conductive medial element is disposed in the gap between the first and second conductive layers, and comprises a second hydrophobic surface. The voltage source communicates with the second conductive layer and the elongate element. By applying a voltage potential between the elongate element and the second conductive layer, droplets can be electrostatically actuated so as to move from the first conductive layer into contact with the second conductive layer. The apparatus is particularly useful in the synthesis of microarrays of biological, chemical, or biochemical samples.
