Abstract: Described are methods, cell growth substrates, and devices that are useful in preparing cell-containing graft materials for administration to patients. Tubular passages can be defined in cell growth substrates to promote distribution of cells into the substrates. Also described are methods and devices for preparing cell-seeded graft compositions, methods and devices for preconditioning cell growth substrates prior to application of cells, and cell seeded grafts having novel substrates, and uses thereof.

Abstract: A graft injector assembly includes a coating disposed on an interior surface of an injector body to form a lumen. The lumen is structured to store a graft comprising corneal tissue and to allow the graft to exit distally from the injector body, preferably with minimal or no damage to the corneal tissue. The corneal tissue may include a Descemet membrane and endothelial cells, which may be used in Descemet Membrane Endothelial Keratoplasty (DMEK). In one example, the assembly includes an organosilane coating disposed on a glass injector body.

Abstract: Engineered tissue compositions for supporting cell growth, maintenance, and/or differentiation featuring a scaffold, extracellular matrix (ECM) material, and optionally a population of ECM-generating cells such as fibroblasts. The tissue compositions may be used for supporting seeded cells of a particular cell type of interest such as cells related to skeletal muscle, smooth muscle, cardiac tissue, gastrointestinal tissue, etc. The tissue compositions with seeded cells may develop into functional tissues, which may have the potential to provide a tissue graft for therapeutic purposes or a valuable model for in vitro assays.

Abstract: A soft tissue device can incorporate a composite material comprising a gel and at least one nanostructure disposed within the gel. A soft tissue device can further incorporate biologically active materials such as cells, tissues. A method for healing a soft tissue defect while promoting soft tissue regeneration can include applying a soft tissue device to a soft tissue defect, wherein the composite material includes a gel and a nanostructure disposed within the gel. A method for manufacturing a soft tissue device for use in healing soft tissue defects can include providing a gel, disposing nanofibers within the gel, and a biologically active material.

Abstract: The invention relates to a cell sheet construct for neurovascular reconstruction. The cell sheet construct has a vascular endothelial cell layer and a neural stem cell layer, and the two layers are physically in direct contact with each other, where the vascular endothelial cell layer forms branching vasculatures, and the neural stem cell layer differentiates into neurons. The invention also relates to a method for manufacturing the cell sheet construct, having the following steps: culturing vascular endothelial cells on a substrate to form a vascular endothelial cell layer, seeding neural stem cells on the vascular endothelial cell layer to make the neural stem cells be physically in direct contact with the vascular endothelial cell layer, and culturing the neural stem cells and the vascular endothelial cell layer to differentiate into neurons and branching vasculatures to form a cell sheet construct.

Abstract: The present invention overcomes all the above drawbacks and provides a versatile method for the fabrication of multilayer hollow tubes that uses a layer-by-layer rod dipping approach using different biomaterials. The device enables fine control over fabrication parameters, such as ascending/descending speeds, rod rotational velocity, and crosslinking or polymerization time. All these technologies allows the generation of more complex multilayer hollow tubes such as vessel-like structures, urethral grafting, prostate grafting and the like.

Abstract: Certain aspects of this disclosure relate to uniquely constructed medical implants that incorporate an electronic or other medical device. Some illustrative pocket-like implants provide an interior space for receipt of an electronic medical device, and are implantable in a patient with the electronic medical device positioned in the interior space. In one form, an inventive construct includes a remodelable material component that is effective upon implantation to promote cellular invasion and ingrowth into the remodelable material so that it becomes replaced by new patient tissue and so that the electronic medical device becomes surrounded by a new pocket structure comprised of newly generated, functional patient tissue. The electronic medical device will be a pacing device or other cardiac rhythm management (CRM) device in select embodiments.
