The present invention relates to a microfluidic system for processing a cell-containing liquid. The system includes a microfabricated substrate having an enrichment channel to prepare an enriched cell sample from the cell-containing liquid. A flow through member is in liquid communication with the enrichment zone. The flow through member substantially prevents cells of the cell-containing fluid from exiting the enrichment zone while allowing liquid of the cell-containing liquid to exit the enrichment zone. A gas actuator associated with the enrichment zone provides a gas pressure sufficient to move the enriched cell sample from the enrichment zone.

The retention member is in communication with the input port and is configured to spatially separate particles of the particle-containing liquidic sample from a first portion of the liquid of the particle containing fluidic sample. The pressure actuator recombines at least some of the separated particles with a subset of the first portion of the liquid separated from the particles. The device can also include a lysing chamber that receives the particles and liquid from the retention member. The lysing chamber thermally lyses the particles to release contents thereof.

Nucleic acid material can be effectively separated from a fluid by first contacting the fluid with a positively charged polymer which binds the nucleic acid material. Thereafter, the polymer, having the nucleic acid material bonded thereto, is contacted with a releasing agent which comprises a solution of an alkaline material and a glycol. The solution has a pH of no more than 12 and operates to release the nucleic acid material from the polymer under relatively low temperature conditions, typically no more than 50° C., and in particular instances, no more than 40° C. The glycol material may comprise a monomeric glycol such as ethylene glycol, propylene glycol, or the like, or it may comprise a polymeric glycol such as polyethylene glycol. Also disclosed is a novel positively charged polymer which may be employed in the separation process. This polymer comprises an acidified polyamine, such as polyethyleneimine which has been reacted with a nonacidified polyethyleneimine in a coupling reaction. The acidified polyethyleneimine may be a coarboxylated and/or sulfonated polyethyleneimine.
