The outlet element and the holding chamber are disposed such that gas flow through the chamber is substantially uniform and unidirectional. A polymer synthesis apparatus (20) for building a polymer chain including a head assembly (21) having an array of nozzles (22) with each nozzle coupled to a reservoir (23) of liquid reagent (24), and a base assembly (25) having an array of reaction wells (26). A transport mechanism (27) aligns the reaction wells (26) and selected nozzles (22) for deposition of the liquid reagent (24) into selected reaction wells (26). A sliding seal (30) is positioned between the head assembly (21) and the base assembly (25) to form a common chamber (31) enclosing both the reaction well (26) and the nozzles (22) therein. A gas inlet (70) into the common chamber (31), upstream from the nozzles (22), and a gas outlet (71) out of the common chamber (31), downstream from the nozzles (22), sweeps the common chamber (31) of toxic fumes emitted by the reagents. Each reaction well (26) includes an orifice (74) extending into the well (26) which is of a size and dimension to form a capillary liquid seal to retain the reagent solution (76) in the well (26) for polymer chain growth therein. A pressure regulating device (82) is provided for controlling a pressure differential, between a first gas pressure exerted on the reaction well (26) and a second gas pressure exerted on an exit (80) of the orifice, such that upon the pressure differential exceeding a predetermined amount, the reagent solution (76) is expelled from the well (26) through the orifice (74). A method of synthesis of a polymer chain in a synthesis apparatus (20) is also included.

Methods for preparing nanoscale reactions using nucleic acids are presented. Nucleic acids are captured saturably, yet reversibly, on the internal surface of the reaction chamber, typically a capillary. Excess nucleic acid is removed and the reaction is performed directly within the capillary. Alternatively, the saturably bound nucleic acid is eluted, dispensing a metered amount of nucleic acid for subsequent reaction in a separate chamber. Devices for effecting the methods of the invention and a system designed advantageously to utilize the methods for high throughput nucleic acid sequencing reactions are also provided.

Disclosed are systems and methods involved in extreme high throughput screening of compounds which have an affinity for a biological target. The system is based on a capillary bundle with two distinguishable ends wherein capillaries on one end are connected to compounds stored in discrete reservoirs and capillaries on the other end are bound and processed to form a two dimensional microarray. A capillary bundle having reaction wells for hybridization and compound reaction in one end of the capillaries is disclosed. The capillaries may be light-conducting. Also disclosed are various methods of identifying a target compound in a liquid using this capillary bundle as well as methods of fabricating the bundle. A novel surface tension guided reaction chamber is also provided. Methods and chemistry for fabrication and use of a surface tension guided reaction chamber in binding and hybridization assays are also disclosed. Methods and systems for precise metering of fluids within the capillaries and at the reaction chambers, including the surface tension guided nullvirtualnull reaction well is provided. Methods for performing high throughput screens using optical fiber lined capillaries of the invention are also provided. The capillary arrays are used with both liquid probes as well as probes immobilized on the walls of the capillary.

The invention relates to methods and systems for high-speed precision dispensing and/or aspirating of microfluidic quantities of reagents and other liquids. The operation of the systems is controlled by data accessed from a customized user-defined text file. Advantageously, the use of such text file control allows high-speed precision dispensing of one or more reagents with a wide dynamic range of dispense volumes in complex combinatorial patterns, ratios and arrays onto or into multiple predetermined locations of a desired target or substrate. This is particularly advantageous when a large number of permutations of different reagent and permutations of reagent volume ratios are involved. The systems may be operated in a high frequency modulated mode to further improve accuracy and reliability.

A liquid from which the droplet is formed contains sample molecules, matrix molecules or a mixture of both sample and matrix molecules. A sample plate is positioned with respect to a position below a portion of the liquid, such as a droplet. A power supply forms an electric field between the sample plate and the liquid, which pulls the droplet to a target location on the sample plate. A plurality of capillary columns is provided for simultaneous or successive multiple depositions via application of one or more high voltage pulses between the liquid sample array and a sample plate.

A system which can analyze compounds with high efficiency, such as in a high throughput drug screening system. The high throughput drug screening system which can test the action of a drug candidate upon a group of cells in a monolayer. A microspace corresponding to a microscopic field area, for example on the order of 100-200 microns in diameter, is isolated from the other cells on the monolayer by creating a seal between a drug delivery perfusion unit and the cells to create the microspace for analysis. A drug candidate is then provided into the isolated microspace. The interaction between the drug candidate and the cells in the isolated microspace can then be evaluated. With the high throughput drug screening system the vast majority of cells on a monolayer can be used for drug testing. The high throughput drug system also makes it more readily available to use primary cells in addition to immortal cells as the cell layer.
